What Happens in the Holding Box at the Trot UP?

I realise that I don't actually know what happens when a horse is directed to the holding box at a trot-up.

Is there any veterinary intervention, hurried examinations, hoof picked out, etc?

The hold vet will have a chat with the rider to determine if any relevant history, they will trot the horse up, have a real good feel, poke, prod etc of everything to determine most likely cause and then chat with the rider about whether they wish to re-present the horse. If yes, the vet will then discuss their findings with the ground jury before the horse is trotted up for them again.

Yes, but it's unlikely to go in their favor as generally it means the Hb vet thinks it's too unsound or has found something they believe will get worse if the continue to the jumping.
